About

La mission de notre laboratoire est de comprendre les bases immunologiques et moléculaires des lymphomes B, avec l’objectif à long terme de développer de nouvelles stratégies thérapeutiques combinant recherche fondamentale, translationnelle et clinique, afin d’améliorer directement la prise en charge des patients.

Le développement des hémopathies lymphoïdes B est le résultat d’un processus complexe et progressif, caractérisé par l’accumulation d’altérations (épi)génétiques et de transformations cellulaires, évoluant dans un microenvironnement spécifique propice à la tumorigenèse. Cette dynamique, qui peut s’étendre sur plusieurs décennies, aboutit à l’émergence de tumeurs hétérogènes, marquées par des réponses thérapeutiques variables et souvent imprévisibles.

Pour répondre à ces enjeux, notre laboratoire s’appuie sur une approche multidisciplinaire et intégrée. Nous mobilisons des technologies de pointe en « multi-omiques » à résolution unicellulaire, des collections biologiques uniques de biopsies issues de cohortes de patients en vie réelle et d’essais cliniques, ainsi que des modèles murins génétiquement modifiés et des outils d’édition génomique innovants fondés sur la technologie CRISPR/Cas9.

Positionné à l’interface entre la recherche biomédicale et la médecine translationnelle, notre laboratoire collabore étroitement avec le Groupe coopérateur menant les essais cliniques sur le lymphome en France (LYSA), l’Institut Carnot Calym à l’interface de la recherche académique et de la recherche partenariale, ainsi qu’avec des partenaires industriels.  A terme, l’intégration de nos découvertes dans le cadre des essais cliniques vise à faire progresser la classification des tumeurs, à développer de nouveaux biomarqueurs prédictifs de la réponse thérapeutique et à ouvrir la voie à des approches thérapeutiques innovantes dans le domaine des lymphomes.

Nos projets de recherche s’articulent autour de trois axes principaux :

  1. Caractériser les cellules précurseurs cancéreuses (CPC) à l’origine des rechutes :  Les lymphomes folliculaires (FL) prennent naissance à partir de cellules précurseurs rares présentes plusieurs années avant le diagnostic mais qui constituent également un réservoir de cellules initiatrices responsables des rechutes après traitement. Ceci suggère que les immuno-chimiothérapies actuelles n’éliminent pas complètement le bulk tumoral laissant persister un petit contingent de CPCs tolérantes au traitement, pour des raisons encore inconnues. En s’appuyant sur des modèles murins précliniques récapitulant la progression indolente de la maladie et des échantillons longitudinaux de patients atteints de LF, nous visons à caractériser de manière fine (phénotypique/ fonctionnelle et génétique) cette population rare de CPC qui représente un enjeu majeur pour mieux traiter et intercepter les rechutes.  Ces avancées devraient permettre la découverte d’options thérapeutiques innovantes ciblant la CPC.
  2.  Comprendre l’hétérogénéité cellulaire des lymphomes B : « Lymphoma Single-Cell Atlas »: En collaboration avec le laboratoire de Pierre Milpied (CIML), nous réalisons des analyses transcriptomiques à la résolution unicellulaire dans les lymphomes. Nous étudions les mécanismes d’hétérogénéité et de plasticité cellulaire qui sous-tendent les propriétés tumorales, cherchant à identifier des programmes transcriptionnels communs susceptibles de servir de biomarqueurs prédictifs de réponse thérapeutique, d’affiner la classification tumorale et d’ouvrir de nouvelles voies thérapeutiques.
  3. Identifier les dépendances oncogéniques et les mécanismes de réponses aux thérapies ciblées dans les lymphomes B : Pour favoriser le développement de nouvelles thérapies et proposer des combinaisons rationnelles, nous déployons des approches de génomique fonctionnelle à haut-débit basées sur les criblages pangénomiques CRISPR/Cas9. Ces approches constituent la base essentielle pour des études mécanistiques et translationnelles futures.
