À propos

Le programme de recherche de l’équipe vise à décrypter les mécanismes moléculaires d’activation des lymphocytes T, un processus central de l’immunité adaptative. Pour ce faire, elle adopte une approche systémique et interdisciplinaire combinant biophysique, biophotonique, protéomique et génie génétique afin d’explorer les dynamiques de signalisation, de la nanométrie à l’échelle de l’organisme entier. L’objectif est de comprendre comment les signaux précoces influencent les décisions cellulaires à court et moyen terme, et ainsi améliorer la conception rationnelle des immunothérapies.

Notre programme s’articule autour de trois axes principaux.

A. Biophysiques de la signalisation transmembranaire (Responsables : HT He & Y Hamon)

  • Physico-chimie membranaire et formation des signalosomes. Lors de l’interaction entre le TCR et le complexe pMHC, la membrane des lymphocytes T subit un remodelage rapide. Ce phénomène favorise la formation de signalosomes, complexes protéiques essentiels à la signalisation. L’équipe utilise des techniques avancées comme la FCS (fluorescence correlation spectroscopy), la microscopie FLIM et des sondes solvatochromiques pour étudier l’ordre lipidique et la tension membranaire, deux paramètres clés influençant la signalisation.
  • Rôle des transporteurs ABC (ABCA1/ABCG1). Ces transporteurs régulent l’efflux du cholestérol et influencent la composition lipidique de la membrane. Leur rôle est étudié dans la régulation de la mécanique membranaire, la configuration du TCR, le trafic intracellulaire et le métabolisme des lymphocytes T. Des modèles murins et des patients humains déficients permettent d’explorer leur impact sur la signalisation et la fonction immunitaire.

B. Biologie intégrative de l’activation des lymphocytes T (Responsables : B Malissen & M Malissen)

  • L’activation des lymphocytes T nécessite à la fois des signaux induits par le récepteur pour l’antigène (TCR) et la molécule de costimulation CD28. La protéine cytosolique CARMIL2 permet l’activation du facteur de transcription NF-kB grâce à sa capacité à lier CD28 à la protéine adaptatrice CARD11. L’équipe a développé des souris exprimant une mutation nommée Carmil2QE qui imite une mutation observée dans les tumeurs malignes des lymphocytes T humains. Les lymphocytes T issus de souris Carmil2QE contiennent des complexes CARMIL2QE-CARD11 préformés en nombre comparable à ceux s’assemblant dans les lymphocytes T après activation de CD28. Ces complexes CARMIL2QE-CARD11 préformés se forment également chez des souris déficientes en CD28. De manière inattendue, ils y induisent la plupart des fonctions normalement liées à l’activation de CD28 et ce de manière antigène dépendante. L’équipe a montré que lorsque des lymphocytes T spécifiques des tumeurs expriment Carmil2QE ils ne nécessitent plus l’activation de CD28 et échappent ainsi à l’inhibition des molécules inhibitrices PD-1 et de CTLA-4. Le rôle primordial joué par les signaux CARMIL2-CARD11 parmi ceux déclenchés par CD28 peut donc être exploités pour induire de puissantes réponses lymphocytaires T spécifiques des tumeurs solides en l’absence de ligands de CD28 et d’inhibiteurs de points de contrôle immunitaires.
  • Maladie liée à IgG4 (IgG4-RD). Cette maladie auto-immune fibro-inflammatoire affecte plusieurs organes. Le modèle souris LatY136F reproduit fidèlement les caractéristiques pathologiques humaines. Grâce à la génétique fonctionnelle et à l’omique, l’équipe explore les mécanismes moléculaires et cellulaires de la maladie, avec l’objectif de développer des thérapies ciblées.

C. Nouvelles technologies pour l’analyse des interactions biomoléculaires (Responsable : D Marguet)

  • Interactions électrodynamiques à longue portée (LEDIs). Le projet européen LINkS explore l’existence d’interactions électrodynamiques à grande distance entre protéines grâce à un consortium interdisciplinaire. Ce projet pourrait révolutionner notre compréhension de l’auto-organisation cellulaire et des effets des champs électromagnétiques sur le vivant.
  • Développement de méthodes FCS, SMLM, polarimétrie. L’équipe perfectionne les techniques de microscopie à molécule unique (SMLM) et de spectroscopie pour observer la dynamique des protéines à haute résolution. L’intégration de la fluorescence polarisée permet d’obtenir des images structurales avec des informations sur l’orientation membranaire.
